Enhancement in corneal permeability of riboflavin using cyclodextrin derivates complexes as a previous step to transepithelial cross-linking

چکیده
Corneal cross-linking has been described as an effective treatment to slow the progression of keratoconus. The standard protocol entails corneal epithelial removal allow diffusion riboflavin into stroma. Although, de-epithelization can generate risks or complications that transepithelial tries solve avoid. Different formulations were developed after verifying hydroxypropyl-?-cyclodextrin (HP?CD) and sulfobuthylether-?-cyclodextrin (SBE?CD) in a 20% concentration, increased solubility practically insoluble water drugs such from 0.12 mg/mL 0.35 0.29 respectively. These values higher when chitosan arginine added formulation, showing 0.78 HP?CD concentration was not modified. Ex vivo permeability measured having kept contact bovine corneas with intact epithelium for 5 h 0.1 solution, reproduced nanoemulsion another work. Riboflavin’s cyclodextrins, chitosan, part formulations, compared control drug solution. best coefficient reached combined 40% (w/v) HP?CD, 0.5% (w/w) arginine, chitosan. After carried out toxicity studies opacity (BCOP) He?s Egg Test - Chorioallantoic Membrane (HET-CAM) it verified both, active ingredients excipients different harmful without generating irritation, loss transparency alterations. results show great potential ocular solution their use keratoconus treatment.

Cross-linking with ultraviolet-a and riboflavin reduces corneal permeability.
PURPOSE To investigate the effect of cross-linking treatment on corneal permeability in a live animal model. METHODS Rabbit eyes were selected at random to be left unoperated or to undergo epithelial debridement with or without treatment consisting of cross-linking (CXL) with riboflavin and ultraviolet-A. Nine eyes received a total dose of 3.6 J/cm² and after epithelial healing the corneas we...
متن کاملEnhancement in corneal permeability of riboflavin using calcium sequestering compounds
Ethylenediaminetetraacetic acid, ethylenediamine-N,N'-disuccinic acid and ethylene glycol-bis(2-aminoethylether)-N,N,N',N'-tetraacetic acid are polyaminocarboxylic acids that are able to sequester metal ions. Calcium is implicated in maintenance of intercellular matrix, zonula occludens (tight junctions) and zonula adherens of epithelium and endothelium cells. Corneal epithelium is impervious t...
متن کاملCorneal Absorption of a New Riboflavin-Nanostructured System for Transepithelial Collagen Cross-Linking
Corneal collagen cross-linking (CXL) has been described as a promising therapy for keratoconus. According to standard CXL protocol, epithelium should be debrided before treatment to allow penetration of riboflavin into the corneal stroma. However, removal of the epithelium can increase procedure risks. In this study we aim to evaluate stromal penetration of a biocompatible riboflavin-based nano...
متن کاملHistopathological Finding of Cornea after Collagen Cross-Linking Using Riboflavin and Ultraviolet A in Rabbit
Purpose: To evaluate the histopathological changes of the cornea in rabbit corneas after accelerated collagen cross-linking. Methods: Right eyes of 7 adult New Zealand albino rabbits weighing 1.5 to 2.0 kg were studied. Right eyes of all animals were in the treatment group and left eyes were in the control group. The epithelium of all right eyes was removed and was crossed-linked with riboflavi...

PURPOSE To report 18-month follow-up after transepithelial cross-linking (CXL) in young patients. METHODS Thirteen eyes with keratoconus were evaluated (mean patient age: 14.4 ± 3.7 years [range: 8 to 18 years]). Corrected distance visual acuity (CDVA); spherical equivalent refraction; keratometry (K); coma, spherical aberration, and higher order aberrations (HOAs) for a 5.0-mm pupil; and thi...
